Sorafenib
Sorafenib (co-developed and co-marketed by Bayer and Onyx Pharmaceuticals as Nexavar), is a kinase inhibitor drug approved for the treatment of primary kidney cancer (advanced renal cell carcinoma), advanced primary liver cancer (]), and radioactive iodine resistant advanced thyroid carcinoma. ==Mechanism of action== Sorafenib is a small molecular inhibitor of several tyrosine protein kinases (VEGFR and PDGFR) (tyrosine kinase inhibitor or TKI) and Raf kinases (more avidly C-Raf than B-Raf). Sorafenib also inhibits some intracellular serine/threonine kinases (e.g. C-Raf, wild-type B-Raf and mutant B-Raf).〔 (See BRAF_(gene)#Sorafenib for details of drug structure interaction with B-Raf.) Sorafenib treatment induces autophagy, which may suppress tumor growth. However, autophagy can also cause drug resistance.
